Studying cryogenic samples at the nanoscale is crucial in life sciences, since the basic elements of life are governed by nanoscale processes. An in-depth understanding of the processes that occur within a cell, tissue or a whole organ, for instance, is impossible without in-depth knowledge of the structures and composition. Cryo-electron tomography (cryo-ET) offers an unprecedented ability to visualize spatial and organizational relationships of biological macromolecules in their native state and environment, including viruses, cells and their organelles, and even tissues and multicellular organisms. However, as a cutting-edge technique in its early stages of development, cryo-ET is also highly complex as it consists of many steps that can be challenging to execute without error even for experienced practitioners.
The major difficulty in cryo-ET is finding the right region of interest in a frozen sample. Our integrated cryo-correlative light and electron microscopy (cryo-CLEM) solution enables in-situ cryo-CLEM to greatly enhance the region of interest (ROI) targeting inside the focused ion beam/scanning electron microscope (FIB/SEM) while reducing the number of handling steps required. To help you keep samples vitrified and ice contamination-free, we developed a series of ice defence tools to minimize the undesired ice growth at every step of your cryo-electron microscopy workflow.
